If queuing with a party after you lock in your 4+ heroes you can click on a party member's name, select view profile, and then back out of their profile and have the ability to freely select what agents you want again even though it still says you are locked in and ready for queue. I did not have a chance to test to see if it will actually let you only select 1 hero this way or not as the game started too quickly to test.
